In vitro study | Phenylbutyrate is a well-known HDAC inhibitor, which increases the gene transcription of some genes and also exerts a therapeutic effect. Phenylbutyrate significantly attenuated MPTP-induced loss of striatal dopamine and loss of tyrosine hydroxylase-positive neurons. In prostate cancer cells, Phenylbutyrate attenuates the Bcl-X of an apoptosis antagonist (L), a DNA-dependent protein kinase for proteins repaired by double-strand breaks, prostate progression marker caveolin-1 and pro-angiogenic vascular endothelial growth factor expression. Phenylbutyrate and ionizing radiation Synergistically Induce Apoptosis in prostate cancer cells. |
In vivo study | Phenylbutyrate significantly prolonged survival and improved clinicopathology and phenotype in transgenic G93AALS mice. Phenylbutyrate ameliorates histone deacetylation and induces expression of nuclear factor-κB(NF-κB) p50, phosphorylated inhibitory subunit, and beta-cell lymphoma 2(BCL-2) in G93A mice, but decreased the expression of cytochrome c and caspase. Phenylbutyrate expresses either the p50 of phosphorylated IκB, the p50 of translocated NF-κB to the nucleus, or the p50 of directly acetylated NF-κB. In a transgenic mouse model of Huntington's disease (HD), Phenylbutyrate increases brain histone acetylation and decreases histone methylation levels as assessed by immunocytochemistry and Western blot. In the striatum, Phenylbutyrate increases mRNA of the ubiquitin-proteasome pathway and down-regulates apoptotic cell death, the immunoreactivity of active caspase 3. |
